FGF23 is expressed primarily in osteoblasts/osteocytes and aberrantly high levels in the circulation acting via signaling through an FGF receptor (FGFR)-Klotho coreceptor complex cause renal phosphate wasting and osteomalacia. We assessed the effects of exogenously added sKL on osteoblasts and bone using Klotho-deficient (kl/kl) mice and cell and organ cultures. sKL induced FGF23 signaling in bone and exacerbated the hypomineralization without exacerbating the hyperphosphatemia, hypercalcemia and hypervitaminosis D in kl/kl mice. The same effects were seen in rodent bone models in vitro, in which we also detected formation of a sKL complex with FGF23-FGFR and decreased Phex (gene responsible for X-linked hypophosphatemic rickets (XLH)/osteomalacia) expression. Further, sKL-FGF23-dependent hypomineralization in vitro was rescued by soluble PHEX. These data suggest that exogenously added sKL directly participates in FGF23 signaling in bone and that PHEX is a downstream effector of the sKL-FGF23-FGFR axis in bone.","subitem_description_language":"en"},{"subitem_description":"This work was supported in part by grants from the Ministry of Education, Science, Sports and Culture, Japan (18592001 and 20592139 to Y Y and 21791788 to T M), and the Canadian Institutes of Health Research (MOP 83704 to J E A).","subitem_description_type":"Other"}]},"item_1617186643794":{"attribute_name":"Publisher","attribute_value_mlt":[{"subitem_publisher":"Society for Endocrinology"}]},"item_1617186702042":{"attribute_name":"Language","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_1617186920753":{"attribute_name":"Source Identifier","attribute_value_mlt":[{"subitem_source_identifier":"0022-0795","subitem_source_identifier_type":"ISSN"},{"subitem_source_identifier":"1479-6805","subitem_source_identifier_type":"ISSN"}]},"item_1617187024783":{"attribute_name":"Page Start","attribute_value_mlt":[{"subitem_start_page":"285"}]},"item_1617187056579":{"attribute_name":"Bibliographic Information","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2018-06","bibliographicIssueDateType":"Issued"},"bibliographicIssueNumber":"3","bibliographicPageEnd":"300 ","bibliographicPageStart":"285","bibliographicVolumeNumber":"237","bibliographic_titles":[{"bibliographic_title":"Journal of Endocrinology "},{"bibliographic_title":"Journal of Endocrinology "}]}]},"item_1617258105262":{"attribute_name":"Resource Type","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_1617265215918":{"attribute_name":"Version Type","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_b1a7d7d4d402bcce","subitem_version_type":"AO"}]},"item_1617353299429":{"attribute_name":"Relation","attribute_value_mlt":[{"subitem_relation_type_id":{"subitem_relation_type_id_text":"10.1530/JOE-17-0683","subitem_relation_type_select":"DOI"}},{"subitem_relation_type_id":{"subitem_relation_type_id_text":"29632215","subitem_relation_type_select":"PMID"}},{"subitem_relation_type_id":{"subitem_relation_type_id_text":"https://doi.org/10.1530/JOE-17-0683","subitem_relation_type_select":"DOI"}}]},"item_1617605131499":{"attribute_name":"File","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_date","date":[{"dateType":"Available","dateValue":"2019-06-01"}],"displaytype":"simple","fileDate":[{"fileDateType":"Available","fileDateValue":"2019-06-01"}],"filename":"JEndocrinol_237_285.pdf","filesize":[{"value":"1.5 MB"}],"mimetype":"application/pdf","url":{"objectType":"fulltext","url":"https://hiroshima.repo.nii.ac.jp/record/2007329/files/JEndocrinol_237_285.pdf"},"version_id":"895b637f-2d75-40ec-87a6-279c94f52e29"}]},"item_1732771732025":{"attribute_name":"旧ID","attribute_value":"49037"},"item_1732772494514":{"attribute_name":"備考","attribute_value":"Post-print version/PDF may be used in an institutional repository after an embargo period of 12 months."},"item_title":"Soluble Klotho causes hypomineralization in Klotho-deficient mice","item_type_id":"40003","owner":"41","path":["1730444907710"],"pubdate":{"attribute_name":"PubDate","attribute_value":"2023-03-18"},"publish_date":"2023-03-18","publish_status":"0","recid":"2007329","relation_version_is_last":true,"title":["Soluble Klotho causes hypomineralization in Klotho-deficient mice"],"weko_creator_id":"41","weko_shared_id":-1},"updated":"2025-02-21T09:44:13.105877+00:00"}
